GENETIC ENGINEERING AND BIOTECHNOLOGY
REPORTER GENE ASSAY
In a reporter gene assay system a chimeric DNA contrast is designed with a gene that codes for a enzyme before the promoter of a gene of interest. the reporters contrast contains an enzyme for easy read-out and eukaryote promoter
LUCIFERASE REPORTER SYSTEM
It is an enzyme found in the abdomen of firefly photinuspyralis. This enzyme
uses D-luciferin as a substrate to form oxyluciferin. The luciferin is converted
into luciferin adenylate in the presence of ATP, Mg2+, by cleaving
pyrophosphate (PPi) and transferring AMP to luciferin. Luciferin adenylate
undergoes oxidative decarboxylation and forms oxyluciferin with the emission
of light.
